Molecular Characteristics of Chicken Liver Arginase By ENRICO GRAZI and ERMES MAGRI
نویسنده
چکیده
A purification procedure for the preparation of chicken liver arginase in a homogeneous form is presented. The enzyme hydrolyses both arginine and argininic acid. Kinetic analysis reveals that the enzyme binds arginine when the amino group is protonated or unprotonated; however, the unprotonated form seems to be hydrolysed more rapidly. The enzyme exchanges with the medium approx. 1.6Mn2+ ions per molecule.

Molecular charcteristics of chicken kidney arginase.
Chicken kidney contains two arginases with different sedimentation coefficients and substrate specificity. The ligher of these arginases, which hydrolyses only L-arginine, has been purified about 3000-fold. Like the "ureotelic" arginase, developed in chicken liver after starvation, it displays many of the properties of the arginase of the "ureotelic" species. This seems to exclude the possibili...
متن کاملSeparating mitochondrial protein assembly and endoplasmic reticulum tethering by selective coupling of Mdm10
The endoplasmic reticulum-mitochondria encounter structure (ERMES) connects the mitochondrial outer membrane with the ER. Multiple functions have been linked to ERMES, including maintenance of mitochondrial morphology, protein assembly and phospholipid homeostasis. Since the mitochondrial distribution and morphology protein Mdm10 is present in both ERMES and the mitochondrial sorting and assemb...
متن کاملThe arginase activity of isolated cell nuclei.
Arginase appears to be one of the first enzymes which was studied in cell nuclei isolated on a relatively large scale by physicochemical methods. This enzyme was measured in isolated nuclei of liver cells by Behrens (1) who reported that the arginase activity per dry weight of isolated nuclei was about equal to the arginase activity of cytoplasm per dry weight in the case of mammalian liver. It...

The release of Pi from the Pi-G-actin-ADP complex is the rate-limiting step in the ATPase activity that is shown by ATP-G-actin in the presence of protamine.
متن کاملMolecular characteristics of rat liver arginase.
The molecular weight of approximately 1,500-fold purified arginase from rat liver (specific activity = 19,500 pmoles of urea per min at 25’ per mg of protein nitrogen) was determined by the method of sedimentation equilibrium to be 118,000. The sedimentation velocity constant is 6.1 S, the diffusion coefficient 5.2 X 10U7 cm2 per sec. Alteration in pH, removal of Mn2+, and replacement of Mn2+ b...
